I too am working on the PIMT project in the Downie lab. My work is two pronged. One aspect is quite long term and parallels work done by Brent (see above). Dr. Downie and I are crossing an Arabidopsis pimt2 mutant David isolated with several lines carrying the GUS reporter gene driven by a constitutive promoter to develop a tool to examine which of several abotic stresses cause the greatest accumulation of isoaspartyl residues (and hence, loss of function) in the reporter. My second project is with David Martin and consists of applying one- and two-dimensional gel electrophoresis followed by western blot to examine the PIMT isoforms present in Arabidopsis plant parts.
